LSR I troubleshooting
- Neither cytometer nor computer will not turn on.
- Make sure that UPS under bench is turned on.
- Computer will not connect to cytometer
- Restart computer
- Cytometer status NOT READY
- Allow to warm up for at least 5 minutes.
- Cytometer status remains at STANDBY
- Fluidics control not set to "RUN"
- Tube is cracked
- Check pressure in sheath tank (Open black tap, depress quick-connect center)
- Check level in sheath tank (pick up tank, compare weight with waste)
- Ensure that sheath tank lid is fitted properly
- Stuck fluidics - turn to STANDBY and back to RUN
- Status READY, but sample will not run
- No cells in sample
- Clumpy sample - filter it
- Blocked SIP or flow cell - PURGE 3 times
- Check pressure in sheath tank (Open black tap, depress quick-connect center)
- Check level in sheath tank (pick up tank, compare weight with waste)
- Ensure that sheath tank lid is fitted properly
- Waste not venting - check waste line
- Air bubbles in sheath filter - purge filter
- Sample runs for a few seconds then stops
- Clumpy sample - filter it
- Blocked SIP or flow cell - PURGE x3
- Waste not venting - check waste line
- Air bubbles in sheath filter - purge filter
- Very slow sample rate
- Sample very dilute
- Fluidics set to LOW
- Fluidics fine control turned down (Note: 10-turn control)
- No signal in FL4/FL5
- UV laser not turned on
- Weak signal in FL4/FL5
- Cells not stained
- Incorrect filter inserted
- PMT voltage too low
- No signal in APC/Cy5 channel
- Pulse processing not turned on
- P8 parameter not set to SSC-W
- Indo-1 signal absent/poor
- UV laser not turned on
- Pulse processing not turned on
- Pulse processing card 2 not set to FL4+FL5
- Check filter in FL4 (should be 405 BP)
- Check filter in FL5 (should be 510 BP)
